PIFT transfers peptide-target interaction patterns to small molecules
Researchers present PIFT, a framework that transfers peptide-target interaction patterns to small-molecule generation for protein-protein interaction targets. It maps peptide-target and small-molecule-target interfaces into a shared target-conditioned interaction space and uses joint sampling-time guidance of masked discrete diffusion and continuous flow matching. A peptide-derived reference steers building-block logits and 3D coordinates while preserving an adapted molecular prior. Experiments across diverse PPI targets report improved interface recovery, docking performance, and favorable molecular properties.
It introduces a concrete cross-modal generative approach for difficult PPI drug targets, relevant to AI-driven drug discovery researchers.
